Neddylation Analysis Service

Neddylation is an important post-translational modification (PTM) in which the small ubiquitin-like protein NEDD8 (Neural precursor cell Expressed Developmentally Downregulated protein 8) is covalently conjugated to lysine residues of substrate proteins through an enzymatic cascade reaction. This process is highly similar to ubiquitination in eukaryotic cells but has unique functional characteristics. The most typical substrates of neddylation are Cullin family proteins. By regulating the Cullin-RING E3 ubiquitin ligase complexes (CRLs), neddylation influences critical biological processes such as cell cycle, DNA repair, signal transduction, and apoptosis.

Studies have shown that abnormal neddylation is closely associated with various diseases, especially tumorigenesis, inflammatory responses, and neurodegenerative disorders. Because of its broad regulatory functions, key molecules in neddylation-related pathways are emerging as potential drug targets. Therefore, systematic neddylation analysis is of great significance for understanding cellular functions and advancing drug discovery.

Services at MtoZ Biolabs

MtoZ Biolabs provides a neddylation analysis service focusing on mass spectrometry detection, enrichment strategies, and immunological validation, forming a complete research system:

1. Target Protein Neddylation Analysis
MtoZ Biolabs offers Target Protein Neddylation Analysis focused on determining whether a specific protein undergoes Neddylation and precisely defining its modification sites. Through immunoprecipitation IP, anti-NEDD8 antibody enrichment, tag-dependent site capture methods, or sequence-specific capture strategies for the protein of interest, MtoZ Biolabs can selectively isolate the target protein from complex samples. Combined with high-resolution LC-MS/MS, we accurately identify lysine residues covalently modified by NEDD8 and support differential analysis across experimental conditions, enabling investigations of protein regulatory mechanisms, ubiquitin ligase system activity changes, and signaling pathway modulation.

2. Neddylation Proteomics Analysis
MtoZ Biolabs provides comprehensive Neddylation Proteomics Analysis, enabling proteome-wide detection and quantitative profiling of Neddylation modifications. By integrating anti-NEDD8 antibody enrichment, K epsilon GG di glycine modified peptide enrichment strategies, chemical tag-based capture approaches, and multidimensional fractionation techniques such as HILIC and SCX, we significantly enhance the detection depth of low-abundance Neddylated peptides. Supported by high resolution LC MS MS with multiple fragmentation modes including HCD and EThcD, this service allows large-scale identification and quantification of Neddylation sites within a single experiment.

Analysis Workflow

1. Sample preparation: Clients provide cell or tissue samples. Proteins are extracted and pretreated for analysis.

2. Enrichment: Modified proteins are enriched using affinity pull-down, antibody immunoprecipitation, or chemical labeling.

3. Digestion and mass spectrometry detection: Samples are enzymatically digested and analyzed by LC-MS/MS with high resolution.

4. Data analysis: Databases and algorithms are applied to identify modification sites and perform quantitative analysis.

5. Result validation: Immunological methods are used to confirm key findings.
